Creation and Phenotypic Analysis of an EMS Mutant Library for Processing Tomatoes
In this study, the seeds of the tomato variety JW001 were subjected to treatment with a 3.5% ethyl methyl methane sulfonate (EMS) solution for 12 h. Following germination and emergence of the seeds, the mutation types and frequencies were counted, and a mutant library of the M2 generation tomato was created. Subsequently, SSR analysis was performed on the characteristic mutations of phenotypic variations. By examining the phenotypic traits of the M2 population, 45 mutant characteristics and 96 mutant individual plants were identified, with a total variation frequency of 14.22%. The specific phenotypic characteristics of the leaves, flowers, fruits, and plants were 57, 22, 63, and 20, accounting for 35.18, 13.58, 38.89, and 12.35% of the overall variation in this study, respectively. The results of SSR analysis revealed that 16 samples exhibited DNA variation. In the M1 and M2 generations, five distinct beneficial mutation types were screened through DNA analysis. This study contributes to the germplasm resources and research foundation for functional genetics and molecular plant breeding of processed tomatoes.
